The enzymatic synthesis of glutamine, a reaction utilizing adenosine triphosphate.
نویسنده
چکیده
The preceding paper (2) describes experiments on the synthesis of glutamine in pigeon liver dispersions, which, together with the results of Krebs and his collaborators (3, 4) and of Bujard and Leuthardt (5), suggested that adenosine triphosphate may play an essential r81e in amide formation. Proof for this function of adenosine triphosphate has been obt.ained in studies with tissue extracts, which are the subject of the present paper. Partially fractionated extracts of acetone-dried pigeon liver have been found to catalyze a stoichiometric reaction between adenosine triphosphate, glutamate, and ammonia, in the presence of magnesium ions, to form glutamine, adenosine diphosphate, and inorganic phosphate.

متن کاملStudies on the Mechanism of Glutamine Synthesis: Evidence for the Formation of Enzyme- bound Activated Glutamic Acid*
Although several enzymatic reactions are now known in which activation of a carboxyl group is associated with cleavage of adenosine triphosphate (ATP) to adenosine diphosphate (ADP) and inorganic phosphate (Pi), the mechanisms of these are not yet understood. Perhaps the most extensively studied enzyme of this group is that which catalyzes the synthesis of glutamine from glutamic acid and ammon...
متن کاملStudies on the Reaction Mechanism of Adenosine Triphosphate : Glutamine Synthetase Adenylyl- transferase from Escherichia coli B
Glutamine establishes feedback control of glutamine synthetase (EC 6.3.1.2) in Escherichia coli over a novel loop, viz. by its effect on an enzymatic adenylylation of glutamine synthetase, whose activity is thus modulated. Positive effecters of the adenylyl transfer reaction, magnesium ion and glutamine, increase the number of -SH groups in adenylyltransferase (EC 2.7.7. -) titratable by 5,5’-d...

Leloir and Cardini in 1953 described an enzyme from Neurospora crassa which formed glucosamine from n-hexose 6-phosphate and n-glutamine (2). Formation of glucosamine by cell-free extracts of streptococci in the presence of glucose, adenosine triphosphate, and glutamine was also reported by Lowther and Rogers (3). Although no similar system was described in mammalian tissues, evidence obtained ...
متن کاملEnzymatic Synthesis of Glutamine in Higher Plants.
The synthesis of glutamine has been studied extensively with enzyme preparations from animal tissues and from bacteria (1, 2, 3, 9, 10). These studies have shown that in such organisms, glutamine may be formed directly from glutamic acid and ammonia, the energy for the synthesis being supplied as adenosine triphosphate (ATP). Little is known, however, of the occurrence or characteristics of sim...
